Interlaboratory comparison studies - Detection (I) Up to and including 2016: ● Feb/March: interlaboratory comparison study on detection of Salmonella in samples from the primary production stage (pps) ● Sept/Oct: interlaboratory comparison study on detection of Salmonella in food or feed samples !! Regularly problems with choice of pps matrix due to Avian Influenza related to migration of wild birds in fall and winter Better to reverse the order of the studies, thus: ● Feb/March: interlaboratory comparison study on detection of Salmonella in food or feed samples ● Sept/Oct: interlaboratory comparison study on detection of Salmonella in samples from the primary production stage 2 Closure workshop May 2017
Interlaboratory comparison studies – Detection (II) Problems when changing the order of the studies: ● Two studies with similar matrix organised closely after one and other ● In one year a study for either pps or food/feed will not be organised Proposed solution: Organise a study with a matrix which can be tested as ‘food sample’ and as ‘pps sample’ Swabs 3 Closure workshop May 2017
Interlaboratory comparison studies – Detection (III) Swabs/sponges used for: ● Sampling after cleaning and disinfection in the primary production stage (ISO 13307) ● Carcass sampling (ISO 17604) ● Sampling of surfaces in the food industry environment and food processing plants (ISO 18593) EURL-Salmonella already performed some experiments (early 2017), by contaminating sponges with a mixture of Salmonella and ‘background flora’ (as possible alternative for chicken faeces) Good (stable) results 4 Closure workshop May 2017
Interlaboratory comparison studies – Detection (IV) Proposal detection studies: ● September/October 2017: Detection of Salmonella in swabs open for NRLs-Salmonella for pps ánd food – use relevant method for own work field (for analysis pps samples or for analysis food). Indicate in reporting form if analysis done as NRL-pps or NRL-food ● February/March 2018: Detection of Salmonella in animal feed ● September/October 2018: Detection of Salmonella in samples from the primary production stage ● February/March 2019: Detection of Salmonella in a food matrix 5 Closure workshop May 2017
Interlaboratory comparison studies - Typing ● Organisation study on typing: October/November 2017 ● For all participants: 20 (+1) serovars for serotyping ● Optional: PFGE for 10 serovars From 2018 (??) replace PFGE or add: (??) ● MLVA typing of Salmonella Typhimurium (and Salmonella Enteritidis?)?? ● Whole genome sequencing (WGS)?? 6 Closure workshop May 2017
Summary results questionnaire MLVA (I) ● Most replies 26/01/2017 – 24/02/2017 ● Sent to all NRLs-Salmonella (36 countries) ● Number of replies (countries): 26 (72%) 1. Do you perform MLVA for non-human isolates? No: 15 (CY, CZ, EE, FI, GR, LV, LT, NO, PL, PT, RO, SK, SI, ES, CH) Yes: 11 (AT, BE, BG, DK, FR, DE, IT, LU, NL, SE, UK) Note: 3 NRLs (EE, FI, NO) mentioned sending isolates to other (human health) institutes for MLVA typing e.g. in case of outbreaks 2. For which Salmonella serovars do you perform MLVA? Salmonella Typhimurium: 11 Salmonella Enteritidis: 8 Salmonella Dublin, Salmonella Derby: 1 7 Closure workshop May 2017
Summary results questionnaire MLVA (II) 3. Where do the isolates come from? Official controls: 9 Outbreak investigations: 9 Research: 9 4. MLVA protocol used for Salmonella Typhimurium? EFSA SOP (2014), incl. set of standardised strains: 4 ECDC SOP (2011), incl. set of standardised strains: 7 5. MLVA protocol used for Salmonella Enteritidis? ECDC SOP (2011), incl. set of standardised strains: 8 6. Number of isolates typed with MLVA in 2016? 0: 1 500-1000: 3 10-100: 4 >1000: 1 (including human isolates) 100-500: 2 8 Closure workshop May 2017
Other activities Work programme 2017/2018 (I) ● Supporting activities: – Test different samples for interlaboratory studies – Ad hoc (e.g. confirmation of typing) – Support in outbreaks (e.g. typing strains) ● Activities for ISO/CEN: – Activities for (draft) standard on PCR for monoSTM – Project leader for development standard on validation of confirmation methods (EN ISO 16140-6) – Project leader for preparing guidance document for drafting standards for microbiology – Analysis results experiments incubation at 35/37 °C and conclude – Preparing draft manuscript validation study CEN mandate – Member of several (other) working groups 9 Closure workshop May 2017
Other activities Work programme 2017/2018 (II) ● Assistance to the Commission: – Ad hoc – Participation in working groups EFSA/DG-Sante – Support in case of outbreaks ● Communication: – Keep EURL website up to date – Newsletter every 3 months – Reporting results in reports (available at website) and when possible in publications 10 Closure workshop May 2017
Other activities Work programme 2017/2018 (III) ● Training – Upon request of NRL (e.g. for molecular typing) or indicated by EURL (in case of poor performance) – Use of BioNumerics, in cooperation with EURLs Listeria and STEC; 4 NRLs per EURL network – Rome, July 2017 (NL 2018) – >2018: WGS? In cooperation with other EURLs (?) ● Molecular typing – Member of EFSA/ECDC steering committee molecular typing data – Curation of data (PFGE) for EFSA database – Curators meetings with EURLs Listeria, STEC and curator ECDC, to harmonise curation of data of EFSA/ECDC molecular database – Organisation interlaboratory studies for molecular typing – Use of WGS especially in outbreaks – Creation of a Working group on WGS of EURLs 11 Closure workshop May 2017

Other subject ● EURL-Listeria monocytogenes (Lm) prepared a guidance document ‘on outsourcing part of proficiency testing trials organised by NRLs for national networks’ ● NRLs-Lm expressed a need for a guidance document, listing: – Criteria to outsource parts of PT schemes organised and supervised by NRLs for national networks; – Criteria to select PT providers, including steps of PT schemes that can be outsourced or not, frequency, details on method used by participants, together with minimum values if possible ● EURL-Lm proposed to make it a general guidance document for all biological EURL networks Need/interest NRLs-Salmonella in a (general) guidance document for outsourcing part of PTs? 13 Closure workshop May 2017
