NUTSHELLS FOR THE DETOXIFICATION OF CHROMIUM(VI) POLLUTED WATER BIOGRAPHY Malvin Moyo holds a BSc. (Honours) Degree in Applied Chemistry (Upper Second-Class Division) from the National University of Science and Technology (Zimbabwe) and an MTech. Degree in Chemistry (Cum Laude) from the Vaal University of Technology. He has, over the past five years, carried out research on the addition of value to waste agricultural materials for development of sustainable water treatment technologies. Malvin has presented his research findings at three conferences and has published three peer-reviewed research articles in reputable academic journals. During the course of his studies, Malvin has co-supervised six students leading to the award of BTech. Degrees in Chemistry. ABSTRACT Driven by the need for sustainably sourced catalysts and the use of reaction systems that generate environmentally benign by-products, the present study aimed to deposit stable, dispersed palladium (Pd) nanoparticles on the modified surfaces of gran- ular Macadamia nutshell biomass for catalytic reduction of noxious hexavalent chromium (Cr(VI) to less toxic trivalent chromi- um (Cr(III)). Through wet impregnation with Pd(II) ions and subsequent hydrazine-mediated reduction to Pd(0), Pd nanoparticles were embedded in a scaffold of polyethyleneimine grafted on bleached Macadamia nutshell biomass previously coated with a chemically bound layer of polyglycidyl methacrylate. Imagery from photography and scanning electron microscopy showed the formation of different layers of the polymeric coating and dispersed palladium resulting from surface modification and palladium embedment, respectively. X-ray diffraction analysis confirmed the formation of pure palladium on the modified MNS surface. The supported nanoparticles exhibited catalytic activity in formic acid-mediated Cr(VI) reduction and showed promising stability with consecutive reuse. He holds a bachelor’s degree and honour in chemistry from Water Sisulu University. He also holds M-Tech degree from Vaal University of Technology. He is enrolled for D-Tech in Chemistry at Vaal University of Technology. During his undergraduate at Walter Sisulu University, he was involved in mentoring and assisting first year with their theory and laboratory chemistry work. During his postgraduate at Vaal University of Technology, he was involved with supervision of B-Tech Chemistry students. He has presented his research findings in many international and national conferences. He published his research articles in peer-reviewed journals. He is currently working as the Technician in Department of Chemistry and also involved in supervision of in-services training students with their Chemistry projects ABSTRACT Nanofibers of poly (methyl methacrylate) (PMMA) incorporated with metal selenide and metal oxide nanoparticles were prepared by electrospinning. The PMMA were coordinated to the metal oxide and metal selenide through carbonyl oxygen atom. The metal oxide and metal selenide have antibacterial and antifungal properties and can improved the thermal stability of PMMA nanofibers. The PMMA incorporated with metal oxide and metal selenide lead to the formation of nanofibers with uneven surface with a diam- eter in the range of 30 to 200 nm. The metal oxide and metal selenide nanoparticles have antibacterial and antifungal activity. These nanoparticles show best activity on gram positive compared to gram positive. Metal oxide nanoparticles show the best activity to bacterial compared to metal sele- nide. All the metal selenide and metal oxide show the best activity against fungi. The nanoparticles were able to inhibit the fungi at lowest concentration. The prepared nanoparticles include: metal oxides (copper oxide, iron oxide, silver oxide), and metal selenide (copper selenide, iron selenide, silver selenide). The prepared metal oxide and metal selenide nanoparticles have good absorption and emission properties consistent with small sizes. These nanoparticles were composed of various phased and stoichiometry de- pending on their compound. In others there were mixture of stoichiometry (metal oxide and metal) and phases. These nanoparticles were dominated by spheres, and some shapes such as rods. The particles size was varying from 1-36 nm depending on the type of nanoparticles. These nanoparticles were prepared using thermal decomposition of a single source (complexes or metal salts). The complexes were prepared from oxygen (urea), and selenium (diphenyldiselenide) ligands. The urea complexes coordinated with metal using oxygen for iron, however in silver complexes both nitrogen and oxygen were used. These complexes allow easy control of reaction parameters, and thermal decomposed to form metal oxide/selenide or metal. The complexes were very stable and decomposed at about 200 °C. INFORMATION SYSTEMS DOCTOR TECHNOLOGIAE: INFORMATION & COMMUNICATION TECH M+6 WAHL Roger Louis THESIS: AN INNOVATIVE INTERNET OF THINGS SOLUTION TO CONTROL REAL-LIFE AUTONOMOUS VEHICLES BIOGRAPHY Roger Wahl has a distinguished and diverse background in the word of technology and currently devotes most of his time to overseeing the complete implementation of big data warehouses for a car dealership. Further to his experience, he was involved in several momentous software and applications development projects in various industries akin to vehicle telematics, insurance and national security and telecommunications. He graduated from VUT in 2009 with a Master’s degree in Information Technology and was accepted into an internship program at Telkom SA and has consequently gained a firm grounding in network engineering and the world of telecommunications. ABSTRACT This research was initiated because of a global increase in congestion on roads and the consequent increase in the rate of fatalities on both national and international roads. Annually, 1.3 million people are killed on the roads globally, and millions are injured. It was estimated that 2.4 million people will be killed in road traffic accidents annually by 2030, and in South Africa, over 14 000 deaths were reported in 2016. The World Health Organisation (WHO) asserted in their 2013 Global Status Safety Report on Road Safety that by 2020, traffic accidents would become the third leading cause of death globally. In this organisation’s 2015 report, South Africa was listed as having one of the highest road fatality rates in the world, averaging 27 out of 100 000 people. Cognisance of these statistics that describe wanton loss of life and serious economic implications, among other reasons, led to the development of autonomous vehicles (AVs), such as Google and Uber’s driverless taxis and Tesla’s autonomous vehicle. Companies have invested in self-driving prototypes, and they bolster this investment with continuous research to rectify imperfections in the technologies and to enable the implementation of AVs on conventional roads. This research aimed to address issues surrounding the systems communication concept, and focused on a novel method of the routing facet of AVs by exploring the mechanisms of the virtual system of packet switching and by applying these same principles to route autonomous vehicles. This implies that automated ve- hicles depart from a source address and arrive at a pre-determined destination address in a manner analogous to packet switching technology in computer networking, where a data packet is allotted a source and destination address as it traverses the Open Systems Interconnection (OSI) model for open system interconnection prior to dissemination through the network. The researcher demonstrated this by designing a computer-simulated Internet of Things (IoT) model of the concept. Experimental research in the form of a computer simulation was adopted as the most appropriate research approach. A prototype was developed containing the algorithms that simulated the theoretical model of IoT vehicular technology. The merits of the constructed prototype were analysed and discussed, and the results obtained from the implementation exercise were shared. Analysis was conducted to verify arguments on assumptions to clarify the theory, and the outcome of the research (an IoT model encompassing vehicular wireless technologies) shows how the basic concept of packet switching can be assimilated as an effective mechanism to route large-scale autonomous vehicles within the IoT milieu, culminating in an effective commuter operating system. DOCTOR TECHNOLOGIAE: BIOTECHNOLOGY M+6 BABURAM Cindy THESIS: ENGINEERING OF AN ENZYME COCKTAIL FOR BIODEGRADATION OF PETROLEUM HYDROCARBONS BASED ON KNOWN ENZYMATIC PATHWAYS AND METAGENOMIC TECHNIQUES BIOGRAPHY Ms C Baburam completed her BSc (Biological Sciences) and BSc Honours (Biological Sciences) at the University of Natal in 2003. She completed an internship at the South African Sugarcane Research Institute (SASRI). Ms C Baburam completed her MSc (Biotechnology) at the University of Stellenbosch in 2008 with bursaries awarded from the National Research Foundation (NRF) and a Sugar Trust Fund Bursary from SASRI. She is currently employed as a lecturer in the Biotechnology Department at the Vaal University of Technology since 2007 and is a researcher in the OMICS Laboratory. She undertook her PhD (Biotechnology) degree in 2016 under the supervision of Dr N.A Feto and was awarded the NRF Thuthuka Grant. Ms Baburam was awarded the Academic Exchange programme in 2017 and 2018. She was also a visiting scientist to the J. Craig Venter Institute (JCVI) in the USA and Macquarie University in Australia to undertake part of her PhD studies. ABSTRACT: ENGINEERING OF AN ENZYME COCKTAIL FOR BIODEGRADATION OF PETROLEUM HYDROCARBONS BASED ON KNOWN ENZYMATIC PATHWAYS AND METAGENOMIC TECHNIQUES Hydrocarbon pollution is a growing environmental concern in South Africa and globally. This inadvertently supports the need to identify enzymes for their targeted degradation. The search for novel biocatalysts have relied on conventional culture-based techniques but this allows sourcing of the biomolecules from only 1-10 % of the microbial population. Hence, the metagenomics approach was implemented. The aim of the study was to extract metagenomic DNA from hydrocarbon contaminated soils and construct a metagenomic fosmid library to be functionally screened for monooxygenases, alcohol dehydrogenases and aldehyde dehydrogenases. The fosmid constructs were sequenced using PacBio next generation sequencing platform. The sequences were de novo assembled and analyzed. Four novel genes were analyzed in silico and codon-optimized. The enzymes were purified and characterized and a cocktail for hydrolysis of hexanol developed. Therefore, novel enzymes were mined from the metagenome of fossil-oil contaminated soil and effective hydrocarbon-degrading enzyme cocktails containing their combination were successfully engineered. MECHANICAL ENGINEERING DOCTOR OF PHILOSOPHY : MECHANICAL ENGINEERING M+6 TCHOMENI KOUEJOU Benard Xavier THESIS: DYNAMIC ANALYSIS OF CRACKED TWIN ROTOR SYSTEM INTERCONNECTED BY A HOOKE’S JOINT IN VISCOUS FLUID ABSTRACT Feature extraction of a specific signal in fluid-structure interaction is among the significant problems in mechanic fields. One of the most complex aspects of fault diagnosis in a mechanical system is signal discretization especially when machine signal interferes with unexpected signals, resulting in the complexity of effective signal recognition. Yet a comprehensive study of such problems remains a challenge due to their high nonlinearity and multidisciplinary nature. The research presented in this dissertation is focused on a particular engineering application of fluid-structure interaction and crack detection in connected twin-rotor. The twin-rotor system was modelled as a continuous rotor whose motion was described by a set of differential equations including the effects of unbalance, transverse crack, motor speed, the hydrodynamic forces and a fluctuating mixed-mode representing the perturbation of the Hooke’s joint. By merging the subsystems, the governing equation of the rotating driveshaft partially surround- ed by an incompressible viscous fluid is established, resulting in a highly coupled seven degree-of-freedom system with strong nonlinear terms. The dynamic analysis of such a complex system concentrated exclusively on the hydrodynamic forces’ influences on a breathing crack mechanism and the transmission of power to a shaft driven by a damaged driveshaft. The first extracted features through the frequency spectrum, orbit patterns and coupled rotor deflection revealed that the complex fluid-driveshaft interaction model has resulted in a comprehensible faults diagnosis analysis when multiple faults coexist in the system. The experimental setup in a filled rectangular container with a GST 32 industrial Oil was then built to validate the numerical results which showed that the performance of the twin-rotor system was significantly impacted by both the perturbation of the Hooke’s joint and the hydrodynamic non-stationary vibration signals sources of multiple frequencies fluctuation. The vibration signals from the twin-rotor system exhibited several abnormalities in the frequency spectrum at the first critical speed due to the presence of noise, limiting the reliability of the methods applied. The time-frequency techniques and the tools based on the Nonlinear Squeezing Time-Frequency Transform (NSTFT) have therefore proven to be suitable for the dynamic analysis of the rotating machine with and without a transverse crack and have demonstrated that both the NSTFT and changes on the 2× harmonic components are robust indicators for crack detection in a fluid medium.
