FACULTY OF EDUCATION 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY TAREK ABDALLA Department: SCHOOL OF SCIENCE AND MATHEMATICS EDUCATION Supervisor: PROF M OGUNNIYI Thesis: The influence of an in-service training programme on Libyan biology teachers’ pedagogical content knowledge (PCK) Description: The study, underpinned by the Shulman’s (1987) theory of Pedagogical Content Knowledge (PCK), used a mixed-methods design to determine the influence of an in-service programme on the instructional practices of a cohort of Libyan Biology teachers. The findings showed that the programme was effective for enhancing the teachers’ cognitive and instructional skills to: implement the new Biology curriculum; organize hands-on activities in Libyan multicultural classrooms. The findings also exemplify Bybee’s (1997) 5Es namely, engaging learners, exploring and explaining phenomena; elaborating scientific concepts and evaluating learners. The findings further showed that the teachers increased their understanding of learners’ needs, sociocultural backgrounds and consequently their interest to study biology. MERVYN AUBREY COETZEE Department: LANGUAGE DEPARTMENT Supervisor: PROF S SIVASUBRAMANIAM Thesis: Trauma, injustice and identity: Investigating an egalitarian and auto-ethnographic approach to analyzing students language narratives Description: The thesis blends the key aspects of life history approach with an autoethnographic epistemic stance to analyze the discourses of affective trauma, injustice and identity in the personal language narratives of the Academic Literacy students enrolled in University of the Western Cape’s Bachelor of Education Degree. The egalitarian lens used in the investigation assumes particular prominence in that it helps examine the students’ real-life worlds via their respective narratives, classroom observations, interviews and survey to come to terms with the relationship between what is expected of them academically and the life-worlds that preceded their entry to the university. In light of this, Mervyn’s personal experience with affective trauma conveyed via his autoethnography lays the groundwork for addressing the issues of affective trauma, injustice and its subsequent impact on students’ identities and self-perception. 12 UNIVERSITY OF THE WESTERN CAPE
FACULTY OF EDUCATION DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY BELINDA MARGARETH CORNELISSEN SCHOOL OF SCIENCE AND MATHEMATICS EDUCATION Supervisor: PROF C JULIE Thesis: Pre-service mathematics teachers’ engagement with the evaluation and construction of alternative mathematical models for the same phenomena Description: The competence of prospective mathematics teachers to evaluate and construct alternative mathematical models should be developed due to mathematical modelling being valued in all school mathematics curricula. Cornelissen investigated issues pertaining to preservice teachers’ evaluation and construction of models dealing with social phenomena. Real-time data were collected. Thematic analysis rendered that fairness was the main concern students had with existing models. Their reconstructed models addressed this issue. For the two different models related to BEE and shareholding by Blacks, the authority of research drove deliberations. The thesis contributes towards the recognition that lived realities structure the evaluation and construction of mathematical models of social issues. FREDUA KWASI-AGYEMAN INSTITUTE FOR POST SCHOOL STUDIES Supervisor: PROF P LANGA Co-supervisor: DR E WOLDEGIORGIS Thesis: Public Funding and student accessibility: A Comparative study of two Public Universities in Africa Description: This study examines the relationship between public funding and student access at the University of the Western Cape and the University of Ghana. Underpinned by resource dependence theory, the study adopts a qualitative methodology via in-depth interview and documentary analysis to gather data to explore the study’s objective. The findings indicate that, the levels of change in public funding have a significant effect on students’ access at the University of the Western Cape. However, the study finds an insignificant relationship between public funding and student access at the University of Ghana. GRADUATION CEREMONY 2021 13
FACULTY OF EDUCATION 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY NTHAMA JERMINA MABOHOPE MATSIE Department: EDUCATIONAL PSYCHOLOGY Supervisor: DR S STOFILE Thesis: Experiences of visually impaired mainstreamed learners and their teachers in an inclusive school in Lesotho Description: Nthama Germina Matsie’s thesis explored the experiences of mainstreamed visually impaired learners in an inclusive school in Lesotho. Matsie’s thesis addresses an area in which there is a great need for high quality data in the Southern African context and provides experiential material which is powerful and instructive. From the perspectives of the participants, practices of their inclusive school system gave them rewarding and detrimental experiences. The symbolic violence of constant exclusion, of denigration of one’s identity, and of relentless exposure to anxiety-provoking situations of being denied access to the curriculum, is an important and central finding in Matsie’s thesis. KONDWANI KELVIN MKANDAWIRE Department: LANGUAGE DEPARTMENT Supervisor: PROF S SIVASUBRAMANIAM Thesis: An investigation of language learning agency in English for Academic Purposes: The Case of the Malawi University of Science and Technology Description: The study while acknowledging the serviceability of English for Academic Purposes, investigates how its one-size fits all teaching and assessment practices ignore the learners’ language learning history and experiences much to the denigration of their agency and voice in the EAP classroom underlying their learning success. Utilizing the strengths of theoretical positions couched in critical pedagogy, ecological affordances and sociocultural positioning, the study argues for an EAP pedagogy that empowers and positions transitioning students for critical learning via their multiple meaning making, agency and voice. The study generates an array of insights for an advancement of critical EAP learning that can engender students’ voice and agency for reading the word and reading the world. It then urges EAP course lecturers to move away from closure-focused EAP practices with a view to augmenting their critical awareness of their world views. 14 UNIVERSITY OF THE WESTERN CAPE
FACULTY OF EDUCATION DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY FLORENCE NDIBUUZA Department: EDUCATIONAL STUDIES Supervisor: PROF P LANGA Co-supervisor: PROF R BISASO Thesis: The academic profession in the rising knowledge societies in Africa: A comparative analysis of Makerere University and the University of the Western Cape Description: This thesis explores the fate of the academic profession in Makerere University in Uganda and the University of the Western Cape in South Africa. Drawing on the neo-institutional theory, a cross-national mixed methods design via survey, semi-structured interviews and document analysis were used to examine the trends in academic research, teaching and community outreach. The results show that institutional reality specific to limited interest and capacity to innovate, enrolment patterns and donor interests unique to each university sometimes overrule state influence in shaping academics priorities. FACULTY OF NATURAL SCIENCES 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY OMOLOLA FOLASADE AFOLAYAN SCHOOL OF PHARMACY Supervisor: PROF D BEUKES Co-supervisor: PROF M TRINDADE Thesis: Discovery of antibacterial lead compounds from marine organisms Description: The rapid emergence of new and antibiotic resistant pathogens together with the slow development of new antibiotics is creating a global health crisis. Ms Afolayan investigated natural compounds produced by marine algae and microorganisms as a potential source of new antibiotics against methicillin resistant Staphylococcus aureus (MRSA), Mycobacterium tuberculosis and Mycobacterium aurum. Using a classical bioassay-guided approach she isolated, elucidated the structures and evaluated the biological activity of the main antimicrobial natural products from the marine algae. In addition, she identified the “hidden” antibiotic potential of the marine bacteria by directly exploring their sequenced genomes and annotating their biosynthetic gene clusters. RAMI MOHAMED YOUSIF AHMED SCHOOL OF PHARMACY Supervisor: PROF H SAMSODIEN Thesis: Development of rifampicin loaded in surface-modified 4.0 G PAMAM dendrimer as a novel anti-tuberculosis pulmonary drug delivery system Description: Nanoparticles, such as polyamidoamine (PAMAM) dendrimers, are synthetic polymeric nanoparticles that have unique features that afford a dendrimer-conjugate complex the possibility to overcome the most common hurdles associated with drug delivery. This study developed a novel pulmonary delivery system for the anti-TB drug rifampicin using surface- modified G4 PAMAM dendrimer nanoparticles, to improve drug solubility, prolong its release, enhance permeability into the macrophages, and decrease the toxicity of the drug-dendrimer conjugates. The in-vitro and ex-vivo data suggests that the developed novel dendrimers are suitable drug carriers in terms of biocompatibility, release behaviour and site-specific delivery. 16 UNIVERSITY OF THE WESTERN CAPE 16
FACULTY OF NATURAL SCIENCES DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY MONIQUE BELINDA BENNETT Department: MEDICAL BIOSCIENCES Supervisor: PROF G VAN DER HORST Co-supervisor: PROF M DE KOCK Thesis: Marine invertebrate sperm: Assessment of sperm quality using computer aided sperm analysis Description: Five species of marine invertebrates or broadcast spawners, were studied. Computer- aided-sperm analysis was used to analyse sperm morphology and sperm motility that play an important role in determining sperm quality that relate to fertilization success. The candidate demonstrated that sperm form played a significant role in sperm hydrodynamics. For example, the faster a sperm of these broadcast spawners swim, the longer is the head length, the thinner is the head width and the shorter the tail length. A helical swimming pattern was a distinctive feature found in all species. However, species-specificity in the diameters and helical pattern was seen. The thesis produces quantitative data that will be of great value in aquaculture and management of the commercial species in the future. RYNO BOTHA Department: PHYSICS AND ASTRONOMY Supervisor: PROF R LINDSAY Co-supervisors: MR C LABUSCHAGNE, DR A WILLIAMS Thesis: Variability and interannual trends in the climatology of radon-222 at two Southern Hemisphere coastal baseline sites Description: This work studied the measurements of the radioactive gas, radon, that were collected at two Southern Hemisphere sites. Radon data can help to understand the large-scale atmospheric circulation patterns that drive events such as extreme droughts and mega-wildfires. Observations were made at Cape Grim in Tasmania and at Cape Point in South Africa. Strongly skewed radon distributions occur, with most events falling into a compact range of low values (from marine air-masses) and a smaller number of events spread over a wide range of high- radon values (continental air-masses). Variations in seasonal radon are evident especially during the period from 2012 to 2017, perhaps associated with the strong El-Niño southern oscillation centred on 2015. The trends seen indicate changes in circulation patterns over time, perhaps associated with climate change. GRADUATION CEREMONY 2021 17
FACULTY OF NATURAL SCIENCES 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY SOULEYMANE DIALLO SOUTH AFRICAN NATIONAL BIOINFORMATICS INSTITUTE Supervisor: PROF A CHRISTOFFELS Co-supervisors: DR D MASIGA, DR M GETAHUN Thesis: Coding of tsetse repellents by olfactory sensory neurons: towards the improvement and development of new novel tsetse repellents Description: Mr Diallo’s thesis investigated olfaction in Glossina fuscipes fuscipes (Tsetse), vector of trypanosomiasis. The study showed that Tsetse is equipped with diverse olfactory sensilla; basiconic, trichoid and coeloconic, and exhibit sexual dimorphism in types and number. Putative receptors for tsetse repellent components and 1-octen-3-ol (attractant odour) were identified using a multidisciplinary approach; behaviour, molecular biology, electrophysiology and bioinformatics. Additionally, using dsRNAi silencing method proved the direct role of Tsetse odorant binding proteins in the detection and perception of 1-octen-3-ol. The study significantly contributes to the understanding of tsetse olfaction and to the development of olfactory based tools for controlling the spread of trypanosomiasis. MONIQUE ENGELBRECHT Department: MEDICAL BIOSCIENCES Supervisor: PROF M DE KOCK Co-supervisor: DR C VANDERVOORDE Thesis: Radiation-induced leukaemia in South Africa: response of lymphocytes and CD34+ cells to different radiation qualities Description: Several epidemiological studies have highlighted that the lifetime risk to develop radiation- induced leukaemia is 3-5 times higher for children compared to adults. The high risk is linked to the high radiosensitivity of the red bone marrow at young age, which harbours the haematopoietic stem and progenitor cells (CD34+ cells). In this thesis, CD34+ cells were isolated from umbilical cord blood and irradiated with photons and fast neutrons. An increased number of initial DNA DSBs was observed after neutron irradiation, as well as higher levels of cytogenetic damage, highlighting the leukemogenic potential of neutrons. A second part of the thesis focussed on the impact of age on radiosensitivity by comparing the DNA damage response of newborn and adult T-lymphocytes. Significant differences were observed between the two types of T-lymphocytes, which can be linked to the immunophenotypic change of T-lymphocytes with age. 18 UNIVERSITY OF THE WESTERN CAPE 18
FACULTY OF NATURAL SCIENCES DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY ABOBAKER SALEH MOHAMED IBRAKAW Department: BIODIVERSITY AND CONSERVATION BIOLOGY Supervisor: PROF J BOATWRIGHT Co-supervisors: PROF A HUSSEIN MOHAMMED, PROF C CUPIDO Thesis: Chemical investigation of some species of Amaryllidaceae from the Greater Cape Region of South Africa as a source of bioactive compounds Description: Amaryllidaceae are known for their content of pharmacologically active alkaloids. The neuroprotective properties of Boophone haemanthoides (BHE), Crossyne flava (CRO), Clivia miniata (CME) and Nerine humilis (NHE) were investigated for their efficacy in the treatment of the neurodegenerative disease Parkinson’s. The chemical investigation of nonpolar constituents of bulbs of BHE yielded thirteen known compounds. The compounds isolated from BHE and CRO showed improved cell viability, increased ATP generation in cells and inhibition of MPP+- induced apoptosis by MPP+. Members of Amaryllidaceae investigated yielded compounds with high neuroprotection potential, and some showed potent cytotoxicity. More studies are required to isolate compounds with a special focus on neuroprotective activity. KHALIED KASKAR Department: MEDICAL BIOSCIENCES Supervisor: PROF R HENKEL Co-supervisor: DR W GIBBONS Thesis: Optimizing embryo culture conditions and spent culture media analysis as predictors of embryo quality and pregnancy Description: Assisted reproduction is a standard medical treatment of infertility. However, the success rate is still relatively low. Khalied Kaskar compared single-step and sequential culture media in different incubators. Single-step media resulted in better blastocyst development. The EmbryoScope showed slight improvements in embryo development. The single step medium in the EmbryoScop is the preferred choice for embryo culture. Insulin or IGF-1 media supplementation or 2%O2 culture did not provide any benefit to embryo development. The C57 mouse strain is more sensitive and may be a better model for quality control assays. ORP in spent media indicates that embryos that developed did not contribute to ROS, but maintained ORP balance. Khalied presented his work at 5 international conferences and a webinar, indicating the high quality of his work. GRADUATION CEREMONY 2021 19
FACULTY OF NATURAL SCIENCES 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY VEDASTUS MAKENE Department: MEDICAL BIOSCIENCES Supervisor: PROF E POOL Thesis: Cell culture biomarkers for monitoring of wastewater pollutants Description: Vedastus did his PhD in an area that is very relevant and important for sustainable livelihoods namely monitoring of pollutants in water. Water pollution has become a major problem and this can have adverse effects on human, animal and environmental health. Vedastus developed a cell culture system to evaluate raw and treated effluents for toxicity and inflammatory activity. He published four articles in peer reviewed, international journals on the data generated from his research and his one examiner commented that his PhD is one of the top 5 % of theses that she has seen. Adding his newly developed methodologies for water monitoring to currently used systems will ensure a more holistic view of the water quality and whether the water can be used for domestic and agricultural use without posing health risks to consumers. NOMPUMELELO THELMA MOBE Department: EARTH SCIENCE Supervisor: PROF D MAZVIMAVI Co-supervisor: DR S DZIKITI Thesis: The influence of canopy cover and cultivar on rates of water use in apple orchards in the Western Cape Province, South Africa Description: This thesis investigated water requirements of selected apple cultivars differ from planting until the trees reach full-bearing age to improve irrigation management. The results from the study showed that canopy size rather than cultivar type was the main driver of water use rates in apple orchards from planting to full-bearing age. Tree transpiration rates were highest in the high canopy cover orchards, followed by the medium canopy orchards, and lastly the low canopy cover orchards. This study has provided an improved understanding of how water use rates of apple cultivars vary with canopy cover. This information assists in developing guidelines on the water requirements for apple orchards. 20 UNIVERSITY OF THE WESTERN CAPE 20
FACULTY OF NATURAL SCIENCES DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY SAEED SEIDU SAIDU MOHAMMED Department: EARTH SCIENCE Supervisor: DR M OPUWARI Co-supervisor: PROF S TITINCHI Thesis: Geochemical Evaluation of Source Rock Potentials and Characterization of Hydrocarbon Occurrences in the Eastern Dahomey Basin, Nigeria Description: Mr. Mohammed’s doctoral studies focused on the petroleum source rocks and oil sand/heavy oil from the Eastern Dahomey Basin, Nigeria. He assessed the organic-rich rocks underlying the oil sand with a multi-parameter method and evaluated the hydrocarbons using different techniques. The source rock assessment revealed the dominance of Type II to Type III kerogen, and the various complementary maturation analysis all showed the source rocks to be thermally immature. The study of oil sand and heavy oil indicated severe widespread biodegradation. His findings advanced our understanding of the petroliferous Afowo Formation and may have applications in exploiting and exploring petroleum in the frontier basin. PORTIA LEAH MOKOENA Department: EARTH SCIENCE Supervisor: DR T KANYERERE Co-supervisors: PROF Y XU, PROF J VAN BEVER DONKER Thesis: Novel approach of using hydrogeochemistry, hydrogeologic and hydrostratigraphic techniques in evaluating coastal aquifers in Heuningnes Catchment, South Africa Description: Coastal groundwater systems store lots of water, but quality of such water remains undesirable. Portia integrated techniques from hydrogeochemistry, hydrogeologic and hydrostratigraphy to provide robust interpretation on groundwater quality. While deep groundwater is fresh and complied to current WHO and SANS241 guidelines, shallow groundwater is saline, hence not suitable for irrigation and domestic uses. Results gave insight for remediation work. Three papers in international journals have been published and one book chapter. GRADUATION CEREMONY 2021 21
FACULTY OF NATURAL SCIENCES 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY ANESU CONRAD MOYO Department: BIOTECHNOLOGY Supervisor: PROF M TRINDADE Thesis: Biological approach to improving the evaporation rates of mine wastewater desalination brine treated in evaporation ponds Description: The disposal of brine, a by-product of mining operations, is a global problem with adverse economic and environmental implications. In South Africa brines are disposed of through evaporation ponds. In this study Anesu investigated the application of pigmented halophilic bacteria as a more environmentally friendly method for increasing the evaporation rates of industrial wastewater brines. A novel pigment-producing Planococcus sp. was isolated from a salt crystallizer pond which improved evaporation rates of mine wastewater by almost 20%. Using genome analyses and chemical mutagenesis, mutants of the strain confirmed that the evaporation rate increase was highest when a C30 carotenoid, which was chemically characterised in this study, was produced by the bacterium. Such a biological approach represents a sustainable option for the mining industry. BELLO LADAN MUHAMMAD Department: PHYSICS AND ASTRONOMY Supervisor: DR F CUMMINGS Thesis: Application of vertically aligned arrays of metal-oxide nanowires in hetero-junction photovoltaics Description: This study was motivated by the global need for clean, renewable energy conversion using simple technologies. Of particular interest is nanotechnology-based photovoltaic (or solar) cells which are cheaper and have remote, off-grid potential compared to common silicon panels. During his research Bello proved that irradiating metal-oxide nanowires with nitrogen plasma effectively improves the ability of the wires to conduct electrons and scatter light- waves. To demonstrate the practical and commercial potential of these findings, the irradiated nanowires were subsequently used to fabricate organic polymer solar cells which exhibited up to 200% improvement in sunlight-to-electricity conversion efficiency compared to devices designed with conventional nanomaterials. 22 UNIVERSITY OF THE WESTERN CAPE 22
FACULTY OF NATURAL SCIENCES DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY FRANCUOIS LLOYD MULLER Department: BIODIVERSITY AND CONSERVATION BIOLOGY Supervisor: PROF J BOATWRIGHT Co-supervisors: PROF L RAITT, DR S CHIMPHNGO, DR M SAMUELS Thesis: Native legume species from the Northern Cape Province of South Africa and their potential as forage crops Description: Using a combination of ecological niche modeling techniques, plant functional traits and indigenous knowledge, this study identified 18 perennial legume species for further evaluation as alternative drought tolerant forages for improved livestock production in water-limited agro-ecological areas. One species, C. sericea, was further screened using GIS techniques and plant eco-physiological measurements for drought tolerance. Results from this study verify the potential of C. sericea as a species with a wide range of adaptive responses to drought allowing it to cope well under drought stress and rapidly recover once the drought is relieved. This makes C. sericea a good candidate for inclusion into forage breeding programs for the development of a fodder crop for water-limited areas. MOSIDI SARAH NHLAPO Department: STATISTICS AND POPULATION STUDIES Supervisor: PROF N STIEGLER Thesis: Assessment of the potential of hospital birth records to estimate the number of births: a case study of Germiston and Nkomazi local municipalities in South-Africa Description: Mosidi Nhalpo embarked three years ago on a very ambitious research topic looking at the quality of births data in South-Africa and at the way to estimate the correct number of births using hospital birth records. Such a research, never attempted before, is of paramount importance for the correct estimates of births, in particular, and for the whole population’s distribution, in general. Mosidi has completed an incredible field work by collecting thousands of births records from numerous hospitals. By doing so, Mosidi has managed to find ways to palliate the problem of incorrect birth registration by showing the importance of hospital records. However, for hospital records to be the source of improvements for birth register, quality of recording must improve in these health facilities. GRADUATION CEREMONY 2021 23
FACULTY OF NATURAL SCIENCES 2021 DOCTOR’S DEGREES DOCTOR OF PHILOSOPHY MBUKENI NKOMO Department: BIOTECHNOLOGY Supervisor: DR A KLEIN Co-supervisor: PROF M KEYSTER Thesis: The role of p-coumaric acid on the physiological and biochemical response of chia seedlings under salt stress Description: The role of phenolic compounds in regulating plant responses under environmental constraints have received considerable attention, albeit not in pseudocereal crops. In this thesis, Mr Mbukeni Nkomo demonstrated the efficacy of p-coumaric acid to enhance chia seedling growth under salt stress by evaluating various physiological and molecular responses. He showed that the p-coumaric acid mediated increase in chia seedling growth under salt stress was via the activation of a ROS-signalling pathway that involves superoxide under the control of proline accumulation. In addition, Mr Nkomo identified key proteins biomarkers that emphasized the role of p-coumaric acid in enhancing plant growth under salt stress conditions. Some of the research data generate in this study was published in a reputable plant science journal with significant impact. SHREE OM Department: COMPUTER SCIENCE Supervisor: PROF W TUCKER Thesis: Voice and rural wireless mesh community networks: a framework to quantify scalability and manage end-user smartphone battery consumption Description: Mr Shree Om produced a scalability framework to deploy wireless mesh community networks in rural low-income regions. The first component of the framework is a scalability quantification model for network engineers to design, deploy, and even re-design any type of wireless mesh community network while offering recommended quality of service to end-users. Since rural users need affordable devices to access such services, the second component of the framework shows how to best configure and use low-cost low-end smartphones. Mr Om compared battery consumption amongst different low-end smartphones in combinations of voice and instant messaging over WiFi, 2G and 3G networks. The battery management model can be applied to any type of smartphone and communication application. FACULTY OF NATURAL SCIENCES DOCTOR’S DEGREES 2021 DOCTOR OF PHILOSOPHY FAZLIN PHEIFFER Department: BIOTECHNOLOGY Supervisor: PROF M TRINDADE Co-supervisor: DR L VAN ZYL Thesis: Investigating the antimicrobial potential of Thalassimonas actinarium Description: Marine invertebrate symbionts produce chemically diverse compounds to protect their host and to establish a competitive advantage. This study identified that T. actiniarum, a marine bacterium that lives within sea anemones, produces antibacterial, anticancer and biofilm inhibitory activity. Chemistry-, genome- and bioactivity-guided approaches were employed to begin to identify the bioactive compounds responsible for these activities, ranging from small metabolites to large protein complexes and exopolysaccharides. This is the first study to demonstrate that T. actiniarum is a promising source of potentially novel pharmaceutically relevant natural products. SOPHIA CATHERINE ROSSOUW SOUTH AFRICAN NATIONAL BIOINFORMATICS INSTITUTE Supervisor: PROF A CHRISTOFFELS Co-supervisor: DR J RIGBY Thesis: Optimisation of proteomics techniques using archival tumour blocks of a South African cohort of colorectal cancer Description: Sophia’s thesis used formalin-fixed (FFPE) human colorectal cancer samples to evaluate protein extraction parameters required for accurate LC-MS/MS analysis. The DRP, SP3/HILIC protein purification methods performed the best across 1, 5 and 10-year-old tumor-containing tissue block ages. Older blocks yielded significantly less protein, and protein extraction with PEG resulted in lower peptide/protein identifications. The results indicate that long-term storage of FFPE tissues does not significantly interfere with retrospective proteomic analysis, and variations in pre-analytical factors spanning a decade, does not affect proteomic signatures to a significant extent. This work adds significantly to decision-making for pathologists who want to use archived tumor samples stored in hospitals. GRADUATION CEREMONY 2021 25

A GIS based modeling approach is adapted to assess the implementation of MAR in terms of suitable sites as well as appropriate scheme, with a case study presented for the West Coast in South Africa. The thesis contributes the original research work in the development of a three-step approach that includes the conceptual model, the GIS-based analysis for selection of suitable sites for MAR supported by numerical modeling.
