Abstracts The Openflexure Microscope Author(s): Elizabeth Ampolini, College of Agriculture and Life Sciences Mentor(s): Jose Ascencio-Ibáñez, College of Agriculture and Life Sciences The Openflexure Microscope is a 3D-printable optical microscope. Using the online schematics, we have been acquiring all the elements necessary to the build up of the microscope. It has been challenging, especially since the 3D printing facilities at NC State were not running during the pandemic. We found a supplier that printed the parts and also found the source for the hardware. All electronics have been ordered and most have arrived. The optical parts are still to be ordered and this is challenging due to the needs of our project, which requires less amplification than the one suggested in the original microscope version. Once all parts will arrive, we will assemble the microscope and use it to analyze Arabidopsis leaf and apical meristem sections stained to detect a geminivirus protein by immunohystochemistry or by fluorescence detection and compare with our original images taken with the fluorescent microscope. This kit will not only allow students to have hands-on experiences with a food science based experiment, but will also allow them to understand more about what is involved in the field of food science. Many current science education kits on the market don’t explain the science aspect of the kit well enough for students to actually gain knowledge of the field at hand. Our kit will not only give students a good understanding of fermentation, but it will also allow for some fun while conducting the experiment. Using our kit, students will be able to get a better understanding of food science through a lab based learning environment that they can do from their home. Since many students have missed out on opportunities to gain lab based knowledge because of COVID-19, a kit like this would allow them to gain these skills even without being in the school house. Coverage of Climate Change in Introductory Biology Textbooks, 1970-2019 Author(s): Rabiya Ansari, College of Agriculture and Life Sciences Mentor(s): Jennifer Landin, College of Sciences Climate change is one of the primary threats to human society, biodiversity, and ecosystem stability. Yet fewer than half of Americans see climate change as a major threat caused by our own actions (Kennedy, 2020). In this study, we use document analysis to quantify textbook coverage of climate change over five decades. We focused on four topics within college-level Introductory Biology texts: 1) amount of coverage (number of sentences), 2) start location of the topic in the textbook, 3) categorization of coverage (description of the greenhouse effect, consequences of global warming, or possible solutions), and 4) the type of data presented in figures. We analyzed 58 textbooks, each with at least 3 editions, to emphasize texts with high market penetration. Our findings show that coverage of climate change increased to 56.9 sentences over time, most of that increase occurring in the 1990s. Our data also indicate that climate change coverage moved backward in books, rather than forward, over time. Approximately half of the coverage, from 1990 to 2019, was devoted to a general description of the greenhouse effect. While the content addressing impacts of climate change increased (21% to 34.2%), sentences dedicated to solutions peaked in the 1990s (8.3%) and then decreased in recent decades (to

To ensure teachers are implementing interventions at high fidelity, coaching strategies have been widely used. However, while previous research on intervention fidelity has described what effective coaching practices are, there is limited research on explicit coaching models. This study was conducted to determine the effectiveness of “COACH”, a specific coaching intervention with the goal of improving the fidelity of an evidence-based fluency intervention. A randomized multiple baseline across participants design was used to measure the effects of the COACH intervention on teachers’ intervention fidelity. Results show a strong pattern between the introduction of the COACH coaching intervention and improved scores on both teacher’s adherence and quality of implementation. The COACH coaching intervention appears to effectively improve teachers’ implementation fidelity, ensuring the effectiveness of the fluency intervention. Effects of hydrolysis and pyrolysis in a bioprocess using pinebark to mitigate VOCs found in swine wastewater Author(s): Victoria Augoustides, College of Engineering Mentor(s): Praveen Kolar, College of Engineering Lucie Guertault, College of Engineering In the agricultural and biological engineering field, the creation of a circular bioeconomy is of high priority and improving upon waste management technologies is a critical part of this greater goal. In NC, the pork industry produces toxic volatile organic compounds (VOCs) at an industrial scale and reducing the cost of treating swine wastewater is of research interest in the BAE department. This research project investigates the potential to create biochar and monosaccharide precursors from a readily available and inexpensive biomass, pine bark. Adding in the production of monosaccharide precursors enables the producer of the biochar to then sell the coproduct and use the revenue to continue producing the biochar. Additionally, the project seeks to compare the effects on the resulting biochar’s adsorption capacity of VOCs from pyrolysis and enzymatic hydrolysis. The present data reflects valuable baseline characterization data related to physical and chemical properties of raw Loblolly pine wood including acid value, the point of zero charge, Time-of- Flight Secondary Ion Mass Spectrometric Analysis, X-Ray Photoelectron Spectroscopic Analysis, and Scanning Electron Microscope-Energy Dispersive Spectroscopic Analysis. A DNS assay confirmed that glucose was able to be recovered from the biomass and biochar has been produced. Subsequent research is being conducted on the adsorption capacity of the biochar as well as the hydrolyzed biomass to remove common VOCs in swine waste, like p- cresol. Developing an Optimized Germ-free Model for Studying the Effect of the Microbiome on Blowfly Developmen Author(s): Ezra Bailey, College of Sciences Mentor(s): Aram Mikaelyan, College of Agriculture and Life Sciences Marissa Cole, College of Agriculture and Life Sciences Blowflies are members of Calliphoridae, a family of flies that exclusively feed on dead flesh. Even though they are in constant interaction with dense microbial communities, little is known about how their interactions with the microbial world regulate their behavior, physiology, and development. Germ-free rearing systems allow the design of experiments with strictly defined microbial communities. Although several protocols have been developed to rear sterile blowflies in large quantities for maggot debridement therapy, these published methods are not ideal for behavioral observations and raising flies axenically or gnotobiotically to adulthood. Our optimized method allows us to axenically raise the blowfly species, Cochliomyia macellaria, and overcomes some of the technical limitations of earlier methods. The standardized protocol uses ethanol for surface sterilization of fly eggs, followed by germ-free rearing of larvae and later adults on autoclaved cat food. The use of autoclavable polypropylene jars fitted with glass lids enables easier observation of the flies through the various developmental stages. By combining reusable components, with an inexpensive, nutritious growth medium, our germ-free rearing system paves the way for exploring the relationship between blowfly life history and their associated microbiomes under controlled conditions. 27
Abstracts High accuracy dataset generation of nanoparticle assembly for machine learning model Author(s): Tiernan Baucom, College of Engineering Mentor(s): Yaroslava Yingling, College of Engineering Akhlak- Ul Mahmood, College of Sciences With research into the emerging topic of nanoengineering becoming increasingly more prevalent in recent years, multifunctional nanoparticles and their properties have become a subject of great interest for their applications in the health industry, manufacturing devices, and consumer products. Some of their proposed applications include usage in drug delivery, medical imaging, and most recently for the COVID-19 vaccine. The properties of nanoparticles are largely determined by the characteristics of the ligands attached to their surface. The ligand length and functional groups can be modified to exhibit desired behaviors in different solutions which is what allows these multifunctional nanoparticles to be utilized for such a wide range of applications. Two nanoparticles of particular interest are gold and iron nanoparticles which contain antimicrobial and magnetic properties respectively. In order to predict their self-assembly properties, we generated a high-quality dataset to train a machine learning model using atomistic Molecular Dynamics simulations with different ligand lengths and chemical functional groups in a variety of solutions. The initial analysis includes study of the root mean squared deviation (RMSD) of atomic positioning, the radial distribution function (RDF), and study of the nanoparticles’ hydrodynamic radius as well as the radius of gyration for seven nanoparticle systems. Going forward, further simulations are planned to analyze the interactions between multiple nanoparticles and on additional nanoparticles systems. The data from these simulations will be incorporated into a database for building machine learning surrogate models to predict nanoparticle assembly and identify the ideal system for a given application. A Review of Knee Ultrasound Image Segmentation Methods to Improve Diagnosis Procedure for Knee Osteoarthritis (KOA) Author(s): Jefferson Bercaw, College of Engineering Mentor(s): Ming Liu, College of Engineering Knee Osteoarthritis (KOA) is an irreversible degenerative disease, therefore early diagnosis and appropriate therapies are important to slow the disease’s progression [1]. An important indicator of the specific stage of KOA is the level of deterioration of the cartilage within the knee, which can be measured quantitatively through the medial cartilage thickness via ultrasound or MRI images. Currently, clinicians use an electronic ruler, and manually delineate the joint space from grayscale medical images, introducing unavoidable variation into their measurements. Through the use of image segmentation methods, the medial cartilage thickness can be calculated automatically from ultrasound images of the knee. In this study, seven existing and emergent image segmentation methods were implemented in MATLAB, including Otsu’s Thresholding, Active-Contour Modeling: Chan-Vese method, Active-Contour Modeling: Edge method, Edge-Detection, k-Means Clustering Segmentation, and Watershed Segmentation, and the Gradient-Vector Flow method. Morphological and mathematical operations were used as needed in order to ensure feasible calculations were collected. Results from each method were analyzed for their statistical interchangeability to a database of current clinical medial thickness calculations derived from clinicians using graphical techniques and basic comparisons of repeatability. After testing a database of 30 ultrasound images and their associated clinically-derived medial thicknesses, there were multiple conclusions drawn about the comparability of each method to the clinical norm. Additional studies are required to test the feasibility of these image processing methodologies as a reliable replacement for manual calculations. 28
